I do not think that you can actually tell if the e-mail is read.

It’s called Open Rate. This statistics is available in all popular email services like eSputnik, MailChimp, GetResponse, UniSender, SendX etc.

Analytics of ESP will show you which recipient opened your letters and how many time did he/she spent for reading. You can also see if they click on your links or not.
