Yes, 12.5.1 (or higher) versions of ASE, SDK, Open Server and Rep Server 12.6 can safely be installed in the same SYBASE directory. You cannot mix and match other versions. To be specific, you cannot install a product using InstallShield into a directory that contains a product that was installed using the older Studio Installer. All products in the directory must be installed using compatible InstallShield versions. For example, if you have a directory that contains both ASE and Replication Server 12.5, you cannot upgrade only the ASE to 12.5.1. Replication Server must also be upgraded to 12.6 using InstallShield at the same time. The Replication Server 12.6 Installation Guide for your platform has more information on this subject.
