How are the pKD mammalian siRNA expression plasmids made and tested?

The DNA oligos are annealed such that they get cloned into the pKD backbone using the SmaI and EcoRI sites. Every pKD plasmid is then tested in tissue culture and its KD ability assessed. We test either by Q-RT-PCR and/or by western blotting.
