For optimal results, the coating antibodies should be left in the wells of the ELISPOT plate for minimal 16 h at 4C. The blocking buffer, detector antibodies and conjugate can be incubated for 1 h at 37C, 2 h at room temperature or overnight at 4C without any problem.
