If you’re going to grow the cells and not just try and cultivate the microorganisms from a cheek cell sample, you shouldn’t use agar at all. Agar gel is used primarily for microorganisms like bacteria, and not for animal cells. Animal cells are grown in medium, normally DMEM (Dulbecco’s Modified Eagle’s Medium) or MEM, depending on the type of cells (sometimes you also have to add other things to optimize the cell growth, and penicillin is often added to prevent bacteria contamination). I’ve never grown cheek cells before, but if you can’t find any good info you can just grow them on the two different mediums and see which one works best.
