Answer this is actually too complex an issue to answer in a forum like this; the plethora of books published on this subject is indicative of this. But in a nutshell it boils down to the following. Muslim radicals/extremists have an antagonistic worldview: distinguishing between true believers (themselves, of course) on the one side, and the unbelievers and ‘hypocrites’ or ‘apostates’ (Muslims not sharing their view) on the other. They genuinely see it as their mission to drive their interpretation of Islam through with any means possible. The anti-American dimension of their agendas is part of that antagonism, and with that they believe to rally support in the Muslim world. To an extent that is due to the onesideness of US policy towards the Muslim world, in particular in the Middle East. US policy since WWII has been unreservedly pro-Israel, and until recently there was little or no acknowledgement of the rights of the Palestinians as a people.
