Cells transfected with siRNA cannot be readily identified. 2) You can have a renewable resource of RNA interference reagent, with plasmid-based shRNA containing a bacterial origin of replication and an antibiotic resistance marker. In contrast, siRNA cannot be replicated and are consumed with each transfection. 3) Plasmid-based RNA interference may be used with virtually any cell line, for stable or transient transfections. However, siRNA may only be used transiently with cell lines with greater than 80 percent transfection efficiencies.
